Abstrak


Phytic acid is the storage form of phosphorus in plant seed and considered as an antinutrient that reduces the bioavailability of some minerals. This compound can bind mineral of P, Na+, Mg+, Ca+, protein and amino acid, so the digestive tract of human and non ruminant animal can not absorb it. Phytase (myo-inositol hexakisphosphate phosphohydrolase) can hydrolyze phytic acid to inorganic phosphate and myo-inositol. The using of phytase for reducing of phytic acid level of food materials can increase the capacity of intestine to absorb minerals, protein and amino acid. The objectives of research are to know the activity and characters of phytase of Aspergillus ficuum produced on solid media and to measure the capacity of phytase to hydrolyze phytic acid on food and feed materials.

The results shown that the activity of phytase from Aspergillus ficuum is 222,17 ugPi/hour/mg protein with purification 63,3 fold. The optimum of temperature, pH, Km and Vmax value of phytase is 55OC, pH 5.0, 0.468 and 0.256 ugPi/hour, respectively. The capacity of phytase to hydrolyze of phytic acid on soybean dan maize powder is 43,36 ugPi/hour/mg protein and 12,76 ugPi/hour/mg protein.
